Endotoxin Removal Performance Data for EndoTrap® HD

Endotoxins (Lipopolysaccharides, LPS) are biologically active structural components of the outer cell membrane of all Gram-negative bacteria. The biological activity of endotoxins is associated with the LPS, toxicity with the lipid component (Lipid A) of LPS and immunogenicity with the polysaccharide components of the molecules. Therefore endotoxins mediate a variety of pro-inflammatory and toxic functions. Hyglos established product range for endotoxin removal, EndoTrap®, produces high yields with extremely low endotoxin levels for maximized research and downstream results. Products are available for large-scale endotoxin removal in biomanufacturing of proteins, antibodies, vaccines, DNA, buffers and cell extracts as well as for small-scale research and development.

A new study evaluates the endotoxin removal efficiency of four endotoxin removal resins from different suppliers, including EndoTrap® HD:

Reich et al. (Univ. Regensburg) studied endotoxin removal and sample recovery rates of four commercially available affinity chromatography resins.

To this end, BSA samples [1 mg/ml] in commonly used buffers (Borate, TRIS, MES and Acetate) containing 15,000 EU/ml E.coli 055:B5 LPS, were purified with each 1 ml of the different resins in gravity flow columns according to the manufacturers’ instructions.

EndoTrap® HD supplied by Hyglos was shown to be the most effective resin for removing endotoxins in comparison to the other resins tested. Furthermore, the purification with the other resins resulted in higher sample loss.
